They roll their window down thinking he has something to tell them (like maybe he noticed a tail light out or something) but then he rolls his window up as theirs comes down. The punch line is basically, "haha, you rolled your window down for no reason." It's silly and pointless, and that's what makes it funny.

Great prank.
It works because the people in the other car think the prankster is going to be a confrontational asshole, then when he puts his own window up, they realise he’s just a merry prankster.
Their laughter is a mixture of genuine appreciation for the clever joke and relief that they aren’t going to get stabbed today.
